Endotoxin Testing in Urine

Urinary tract infection

The pathogenic bacteria of urinary tract infections are mostly G-bacteria. The levels of endotoxin in normal urine and infected urine are very different, and the testing of urine endotoxin content is consistent with the results of bacterial culture of urine. It has important significance. Endotoxin is the main component of the cell wall of Gram-negative bacteria, which can cause fever, microcirculation disorders, endotoxin shock, and disseminated intravascular coagulation. It can only be released after the bacteria die. Urinary tract infection is one of the common diseases of the urinary system. It is a disease caused by the proliferation of a large number of microorganisms in the urethra. If not treated in time, it will cause chronic infection. Gram-negative bacteria are the main bacteria causing urinary tract infections. As a gold standard for diagnosing urinary tract infections, urinary bacterial cultures can provide clinical observations of the number of clinically infected bacteria, drug sensitivity, and corresponding efficacy. However, urinary bacterial culture takes about 2 to 4 days, which cannot provide clinical treatment basis in time. The testing of urine endotoxin can prompt the infection of Gram-negative bacteria, and can provide clinicians with reference information for diagnosis and treatment efficacy in a timely manner. Foreign reports indicate that when treated with antibacterial drugs, urine culture bacteria may not grow, but endotoxin is positive. Because endotoxins are released after bacterial death during antimicrobial treatment, the determination of endotoxin may be more sensitive than bacterial culture.

Method

Creative BioMart uses Limulus Amebocyte Lysate test for urine endotoxin testing. LAL test is a kind of arthropod. Amebocytes in the blood contain factor C, factor B and proclottin-gEnzyme, which can be activated by endotoxin. The activated coagulase can make the coagulation in the deformed cells coagulagen is coagulated to form coagulated proteins, which can be used for qualitative, semi-quantitative and quantitative determination of endotoxin. The reaction has high sensitivity (pg/ml) and specificity. It can be divided into gel method, turbidity method and color matrix method.

Endotoxin testing container and sample preparation

In order to eliminate the contamination of exogenous endotoxin, the containers and samples used for detecting endotoxin in body fluids need to be pretreated. The utensils used should be baked at 250 ° C for at least 2 hours to inactivate exogenous endotoxin. The test sample was urine. When the LAL test is used to detect bacterial endotoxins, there is no interference with urine, so no special treatment is required. Samples are usually collected by a sterile, pyrogen-free method and placed in sterile, pyrogen-free test tubes. If the sample is cloudy, the supernatant is centrifuged.
