High concentrations of endotoxin are released into the lysate during cell lysis. Endotoxins are negatively charged and can be purified with DNA in both silica-based and anion-exchange-based systems. Typically, endotoxin monomers are between 10 and 20 kDa, but sometimes aggregates with molecular weights greater than 1000 kDa are formed. Therefore, standard systems for plasmid purification without special handling may result in high levels of endotoxin contamination.
