Endotoxin Testing in Serum

Bacterial endotoxin in serum is closely related to many diseases, and endotoxemia is mostly related to the deterioration of the disease, and it decreases with the remission of the disease. Therefore, bacterial endotoxin can be used as an important reference index for measuring the condition and judging the prognosis. Endotoxemia can occur when a large number of Gram-negative pathogens die in the lesion or blood stream, and a large amount of endotoxin is released into the blood. A large number of endotoxins affect the body's macrophages, neutrophils, endothelial cells, platelets, as well as the complement system and the coagulation system, etc., which will produce interleukins 1, 6, 8 and tumor necrosis factor α, histamine, 5 Bioactive substances such as serotonin, prostaglandin, kinin. These substances act on small blood vessels and cause functional disturbances that lead to microcirculation disorders. Clinical manifestations include microcirculation failure, hypotension, hypoxia, acidosis, etc., which leads to shock. This pathological reaction is called endotoxin shock.

Application of bacterial endotoxin in various departments:

  1. Respiratory department: acute respiratory distress syndrome, pneumonia, chronic branch.
  2. Burn Department: Burn infection.
  3. Gastroenterology: intestinal endotoxemia, hepatobiliary disease, acute pancreatitis.

In addition to the above, it is helpful for departments such as infectious diseases, oncology, neurology (surgery), and heart medicine.

Method

Creative BioMart's serum endotoxin test mainly uses Turbidimetric LAL Assay method, which is aims to detect endotoxin by changing the absorbance caused by the formation of a gel by LAL and endotoxin interactions. The mechanism of this reaction is a complex enzyme-linked reaction, as shown in the figure below. In the presence of LPS, factor C in LAL/TAL is activated, which activates factor B. Factor B activates the proclotting enzyme to clotting enzyme. Finally, the clotting enzyme cleaves the coagulogen into coagulin, which binds together to form a clot. The process of gel formation causes the solution to become cloudy and cause changes in absorbance, and we use this to determine the presence or absence of endotoxin.
